New Study: KRysin™ Delivers Near-Complete Digestion for Deeper Plasma Proteomics and Phosphoproteomics
Following the launch of KRysin at HUPO 2026, the full study by Batth et al. is now available! The study is a collaboration between Genovis and the University of Copenhagen. It presents a systematic characterization of KRysin, a high-fidelity protease for bottom-up proteomic analysis of human plasma and enriched phosphopeptides.
Where Trypsin Falls Short
Even after overnight digestion, Trypsin leaves missed cleavages, particularly when lysine or arginine is followed by proline, flanked by acidic residues, or located close to phosphorylated residues. Missed cleavages inflate the search space and split the signal for one sequence across several peptide forms, which lowers proteome depth and reproducibility. KRysin generates tryptic peptides compatible with standard workflows, but it digests proteins through a catalytic mechanism distinct from that of Trypsin.
Same Specificity, Fewer Missed Cleavages
In HeLa digests, 98.8% of KRysin peptides ended in arginine or lysine, including at sites followed by proline. With KRysin, 93.9% of peptides had no missed cleavages, compared with 82.7% for Trypsin/Lys-C and 70.1% for Trypsin. KRysin also remained active in up to 3 M urea.
More Phosphosites From a Third of the Enzyme
KRysin identified 49,888 class I phosphosites per sample, more than 10,000 more than Trypsin and Trypsin/Lys-C. With KRysin, 80% of phosphopeptides were fully cleaved, compared with 57.5% for Trypsin/Lys-C and 51% for Trypsin. Notably, KRysin at a 1:150 enzyme-to-protein ratio quantified more phosphosites than Trypsin/Lys-C at 1:50, using one third of the enzyme.
Three Times the Throughput in Plasma
Plasma samples were analyzed using data-independent acquisition LC/MS/MS at throughputs from 60 to 500 samples per day. KRysin identified 31 to 40% more protein groups than Trypsin. KRysin at 200 samples per day matched Trypsin at 60 samples per day, giving three times higher throughput at the same depth. The protein groups identified only with KRysin were enriched for receptors, adhesion molecules, cytokines and growth factors. KRysin also reached close to its maximum depth after just 1 to 2 hours of digestion, whereas Trypsin/Lys-C required overnight digestion in the direct workflow.
In Summary
KRysin combines the familiar specificity of Trypsin with a much more complete digest. It delivers deeper phosphoproteome and plasma proteome coverage, shorter digestion times and higher throughput, and it fits directly into existing proteomics pipelines.
