FabRICATOR Preclinical Grade (IdeS) is an IgG-specific cysteine protease with validated low endotoxin levels, developed for in vivo preclinical research into IgG-mediated disease and immunomodulation. It digests human IgG at a single site below the hinge, generating homogeneous F(ab’)2 and Fc fragments for rapid reduction of pathogenic or neutralizing IgG in gene therapy, autoimmunity, and transplantation research models.

FabRICATOR Preclinical Grade is a cysteine protease that digests human IgG at a single site below the hinge, generating a homogeneous pool of F(ab’)2 and scFc fragments. IgG is the only known substrate, which makes FabRICATOR suitable for use in complex samples and in vivo.
In vivo IgG digestion has emerged as a powerful approach to modulate humoral immunity. Digested IgG is rapidly cleared from the circulation, selectively inhibiting IgG-mediated immune functions. Applications range from removing pre-existing anti-AAV neutralizing antibodies prior to gene therapy dosing to preclinical research in autoimmunity and transplantation.
FabRICATOR Preclinical Grade is manufactured with validated and controlled low endotoxin levels, making it suitable for a wide range of in vivo preclinical studies. The enzyme is cloned from Streptococcus pyogenes and recombinantly expressed in E. coli. It carries a His-tag and has a molecular weight of 38 kDa.
