FabRICATOR® Z Preclinical – Below Hinge Digestion of Mouse IgG
FabRICATOR Z (IdeZ) is a cysteine protease that digests mouse IgG2a and IgG3 at a specific site below the hinge, generating a homogenous pool of F(ab’)2 and Fc fragments after a two-hour incubation.

FabRICATOR Z Preclinical Grade is an IgG-specific cysteine protease with validated low endotoxin levels, developed for in vivo preclinical research into IgG-mediated disease and immunomodulation. It digests IgG at a single site below the hinge, generating homogeneous F(ab’)2 and Fc fragments, and is more efficient than IdeS (FabRICATOR) on mouse IgG2a and IgG3 – making it the preferred enzyme for murine models in gene therapy, autoimmunity, and transplantation research.

About FabRICATOR® Z Preclinical
FabRICATOR Z Preclinical Grade is a cysteine protease that digests IgG at a single site below the hinge, generating a homogeneous pool of F(ab’)2 and scFc fragments. Compared with IdeS (FabRICATOR), FabRICATOR Z is markedly more efficient digesting mouse IgG2a and IgG3, and it is also active on human, monkey, rabbit, and sheep IgG. IgG is the only known substrate, which makes FabRICATOR Z suitable for use in complex samples and in vivo.
In vivo IgG digestion has emerged as a powerful approach to modulate humoral immunity. Digested IgG is rapidly cleared from the circulation, selectively inhibiting IgG-mediated immune functions. Its higher efficiency on mouse IgG makes FabRICATOR Z the preferred choice for murine disease models.
FabRICATOR Z Preclinical Grade is manufactured with validated and controlled low endotoxin levels, making it suitable for a wide range of in vivo preclinical studies. The enzyme is cloned from Streptococcus equi subsp. zooepidemicus and recombinantly expressed in E. coli. It carries a His-tag and has a molecular weight of 36 kDa.
