GenovisWebinar - Ultrafast Microdroplet Digestion of Antibodies with Fc Silencing Mutations

FabRICATOR® Xtra LALA is an IgG-specific protease that digests below the hinge of antibodies which have been engineered to contain mutated hinges. While traditional in-solution digestion typically requires 30 minutes to several hours, microdroplet digestion enables efficient subunit generation in less than 1 millisecond during spray ionization.

Combined with flow injection analysis (FIA) and online high-resolution mass spectrometry, the method delivers a total analysis cycle time of just 1 minute per sample. For highly digestion-resistant antibodies such as PD-L1 (LALA-PG mutated), this represents a digestion speed up to 9 million times faster than conventional approaches.

We are excited to welcome Yongqing Yang from New Jersey Institute of Technology who will share how this rapid, robust and scalable workflow enables high-throughput screening and characterization of engineered therapeutic antibodies, with 94 samples analyzed in just 104 minutes.
