LC-MS is an important technique for mAb characterization and the monitoring of potentially important attributes and modifications. Middle-level analysis is an efficient way to characterize and monitor your mAbs as it allows for much more detailed modification information to be determined compared to an intact approach. Overall efficiency of these approaches can be improved further by automating the analytical process enabling detailed middle-level characterization to be performed without the requirement for any hands-on sample preparation.
 
We are delighted to invite Claire Butré from Quality Assistance to describe how using FabRICATOR® HPLC columns has enabled the development of fully automated (2D-)LC-MS workflows for multi-attribute monitoring (MAM) analysis of mAbs at the subunit level in a QC-compliant manner.
 
Approaches like this allow for detailed mAb characterization without the need for manual sample preparation, which can make overall process optimization both faster and easier!

FabRICATOR (IdeS) is an IgG-specific cysteine protease that digests antibodies at a single amino acid site below the hinge, generating homogenous F(ab’)2 and Fc fragments.
