Broad immunoglobulin capture across multiple isotypes using Fab-targeted binding for simplified, immuno-profiling workflows.

Conventional immunoaffinity reagents are often limited to selected antibody classes, restricting access to the full immunoglobulin repertoire present in biological samples. AlligBAITOR efficiently captures a wide range of immunoglobulin isotypes, increasing the depth of relevant information that can be derived from complex samples. Here, we describe how AlligBAITOR enables rapid and high-affinity capture of multiple immunoglobulin isotypes, extending its applicability beyond more traditional, IgG-focused workflows.

AlligBAITOR binds antibody Fab domains with high affinity, enabling efficient interaction with diverse immunoglobulin classes in just 30 minutes. Size-exclusion chromatography (SEC) analysis demonstrates complex formation between AlligBAITOR and multiple immunoglobulin isotypes, confirming broad isotype recognition and supporting the use of AlligBAITOR for applications dependent on pan-Ig capture.  Such applications include autoantibody profiling for unbiased biomarker discovery, therapeutic drug monitoring, anti-drug antibody (ADA) immunogenicity assessment in biologics development, infectious disease serology; vaccine response profiling, and population-scale immune surveillance.

AlligBAITOR Lyophilized binds all immunoglobulin isotypes. a) IVIg, b) serum IgA and c) serum IgE were incubated with AlligBAITOR Lyophilized for 30 min at RT. Samples were analyzed by size-exclusion chromatography (SEC) using a sodium phosphate buffer (pH 7.0) on an Acquity Premier Protein SEC 250Å column. Chromatograms for each sample are shown in the absence (upper panels) and presence (lower panels) of AlligBAITOR. Incubation with AlligBAITOR resulted in a shift toward shorter retention times, consistent with the formation of antibody-AlligBAITOR complexes, demonstrating that AlligBAITOR interacts with IgG, IgA, and IgE.
