Lyophilized enzyme for above hinge digestion of human IgA1 and IgA2m1

IgASAP is a family of IgA-digesting enzymes. IgASAP Sub1 is an IgA1-specific enzyme that digests human IgA1 at one specific site above the hinge, and IgASAP Sub1+2 specifically digests IgA1 and IgA2m1 above the hinge. Both enzymes generate intact and homogenous Fab and Fc fragments.
Since the hinge of human IgA1 is longer than that of IgA2, the amino acids C-terminally from the digestion site differs between IgA1 and IgA2m1. IgASAP Sub1+2 digests both secretory and serum IgA. The incubation time is overnight (16-18 h), and due to the specificity of the enzyme, there is no risk of overdigestion. The enzyme is active at pH 6.0 to 8.0 and does not require reducing conditions or co-factors for activity. The optimal activity is at 37°C. IgASAP Sub1+2 is cloned from Erysipelatoclostridium ramosum and expressed in E. coli. The enzyme contains a His-tag and has a molecular weight of 131 kDa.
IgASAP Sub1+2 Lyophilized is available as a lyophilized powder in 1000 unit vials for digestion of 1 mg IgA1 and IgA2m1.

One unit IgASAP Sub1+2 digests ≥ 90% of 1 µg human IgA of IgA1 and IgA2m1 type when incubated in PBS (10 mM sodium phosphate, 140 mM NaCl, 2.7 mM KCl), pH 7.4 at 37°C overnight (16-18 hours).
IgASAP Sub1+2 Lyophilized is reconstituted by the addition of water. No preservatives added. After reconstitution, the enzyme is stable for 1 month at +4-8°C. The product is shipped at ambient temperature, and the content should be stored at -20°C upon arrival.

There is no known enzyme that targets IgA2-only since the amino acids in the hinge regions of IgA2 is present also in IgA1. However, IgASAP Sub1 will digest IgA1-only.
IgASAP Sub1 specifically targets IgA1 and exhibits a faster reaction time (1 hour) compared to IgASAP Sub1+2 (16-18 hours). The cleavage site of IgASAP Sub1+2 is located six amino acids above that of IgASAP Sub1, resulting in the absence of potentially O-glycosylated residues on the Fab fragments. This characteristic can be advantageous in certain cases.
