IgASAP™ Sub1 Lyophilized

Lyophilized enzyme for above hinge digestion of human IgA1

Available Products

IgASAP is a family of IgA-digesting enzymes. IgASAP Sub1 is an IgA1-specific enzyme that digests human IgA1 at one specific site above the hinge, and IgASAP Sub1+2 specifically digests IgA1 and IgA2m1 above the hinge. Both enzymes generate intact and homogenous Fab and Fc fragments.

IgASAP Sub1 hydrolyzes both secretory and serum IgA1. The digestion is complete within 1 hour, and due to the specificity of the enzyme, there is no risk of overdigestion if the incubation time is prolonged. The enzyme is active at pH 6.5 to 8.0 and does not require reducing conditions or co-factors for activity. The optimal activity is at 37°C, but the digestion can also be performed at room temperature using an increased incubation time.
IgASAP Sub1 is cloned from Streptococcus oralis and expressed in E. coli. The enzyme contains a His-tag and has a molecular weight of 148 kDa.
IgASAP Sub1 Lyophilized is available as a lyophilized powder in 1000 unit vials for digestion of 1 mg IgA1.

Product Information

Unit Defintion

One unit IgASAP Sub1 digests ≥ 90% of 1 µg human IgA1 when incubated in PBS (10 mM sodium phosphate, 140 mM NaCl, 2.7 mM KCl), pH 7.4 at 37°C for 1 hour.

Content and Storage

IgASAP Sub1 Lyophilized is reconstituted by the addition of water. No preservatives added. After reconstitution, the enzyme is stable for 1 month at +4-8°C. The product is shipped at ambient temperature, and the content should be stored at -20°C upon arrival.

FAQ and Support

The enzyme is active at pH 6.5 to 8.0, tolerates up to 300 mM NaCl, and does not require reducing conditions or co-factors for activity. PBS and TBS buffers are both compatible. Optimization may be required if buffers other than the recommended are used.

IgASAP Sub1 may digest IgA1 from other primate species with similar hinge sequences, but this has not been tested. IgASAP Sub1 does not digest IgA2.

The optimal activity is at 37°C, but the digestion can also be performed at room temperature using a prolonged incubation time.
