Immobilized enzyme on magnetic beads for automated digestion of IgG below the hinge

FabRICATOR (IdeS) is an IgG-specific cysteine protease that digests antibodies at a single amino acid site below the hinge, generating homogenous F(ab’)2 and Fc fragments.
The FabRICATOR MagIC magnetic beads contains FabRICATOR Immobilized enzyme. The beads enable fast automated digestion of multiple antibodies in parallel in a middle-level characterization workflow. They are designed for automation platforms, but perform equally well on a shaking table if automation is not available.
After digestion, the samples can be directly transferred to the autosampler of the LC-MS for fast chromatography or to another analytical method for analysis. Automated parallel antibody digestion and subunit generation allow for the processing of the numerous samples associated with clone selection, process development and stability studies for a potential biopharmaceutical. Automated antibody subunit analysis reduces the operator time and the risk for sample handling errors, and also increases the throughput.

Sample size: Digestion of up to 24 x 200 µg IgG (2ml) and 96 x 200 µg IgG (4 x 2 ml)
Short Specification: 20µL settled magnetic agarose resin digest >=95% of 20 and 200µg human IgG1 in 100µL 10 mM Sodium Phosphate, 150 mM NaCl pH 7.4 in 20 minutes at 37°C as monitored by SDS-PAGE

0.05% Polysorbate 20 in the digestion buffer to minimize shear stress in automated systems.
The performance of FabRICATOR MagIC at 37°C is typically less than 20 minute digestion with efficiency of >95% for 20-200 µg human IgG1 (1 mg/ml) sample in PBS, pH 7.4.
FabRICATOR MagIC is supplied as a 25% bulk resin in vials. The product is shipped cold and should be stored at +4-8°C.

We can only guarantee optimal digestion for one-time use. We do not have a cleaning or regeneration protocol to provide. However, depending on the antibody and the following application the column can be reused. We do not recommend using with different antibodies due to the risk for contamination of carry-over from previous sample. If column reuse is desired store the column in 10-20% ethanol at +4-8°C.
FabRICATOR is a cysteine protease. It should not result in any other modifications than the digestion of the peptide bond. Digestion will be between the two Glycines (…CPAPELLG / GPSVF…) and the C-terminus will regain the carboxyl group – that is addition of one OH-group and the N-terminal amino group will be regained (addition of one H).
In summary, R1-CO-NH-R2 + H2O –> R1-COOH + NH2-R2.
