SEQURNA Thermostable RNase Inhibitor workflow

SEQURNA Thermostable RNase Inhibitor consists of a proprietary mix of synthetic molecules that protects RNA by interfering with the active site of RNases. It is the ideal choice in library preparation for single-cell RNA sequencing, in situ RNA sequencing, multiomics applications, or other methods that rely on consistent RNase inhibition.

The SEQURNA RNase Inhibitor is thermostable, enduring 55°C for 24 hours or 70°C for 2 hours, and can tolerate even harsher experimental conditions. This ensures robust RNA protection throughout and after heat cycles, outperforming commonly used protein-based recombinant RNase inhibitors. The stability of the inhibitor enables the development of new workflows and enhances RNA integrity over longer experimental timelines, as well as during sample handling and storage.

SEQURNA Thermostable RNase Inhibitor tolerates freeze/thaw cycles and vigorous vortexing, requires no DTT, and remains active across a broad pH range of 4.0-10.0.

Detailed SmartSeq Protocols

In these modified Smart-seq protocols, SEQURNA Thermostable RNase Inhibitor replaces the recombinant RNase inhibitor used in the original protocols. The key difference is that SEQURNA is added only to the cell lysis buffer (the cell collection buffer) and not reintroduced during Reverse Transcription. Unlike the protein-based inhibitor, which loses activity at elevated temperature, SEQURNA remains effective throughout the 72°C cell lysis and RNA denaturation step, as well as the RT reaction.

Unit Definition

One unit SEQURNA Thermostable RNase Inhibitor is defined as a mass unit. 5 mass units/µl inhibits 0.1 pg of RNase A or RNase B in a 10 µl reaction (200 ng ssRNA) by ≥ 50% when incubated at 37°C for 1 hour.

SEQURNA Thermostable RNase Inhibitor is supplied as an aqueous solution with a concentration of 50 mass units/ul. The product is shipped at ambient temperature, and should be stored at room temperature (15-30°C) upon arrival. It also tolerates deep freezing.
