Digestion of IgM

IgMBRAZOR is a unique IgM-specific protease that digests human IgM at one specific site below the CH2 domain in the heavy chain, generating homogeneous F(ab’)2 and Fc fragments.

The enzyme enables middle-level analysis of the complex and high molecular weight human IgM, which facilitates IgM characterization during, for example, the development of vaccines, therapeutics, and diagnostics.

About IgMBRAZOR™

IgMBRAZOR is a unique IgM-specific enzyme that digests human IgM at one specific site below the CH2 domain in the heavy chain, generating F(ab’)2 and Fc fragments.

The digestion is complete within 30 minutes, and due to the specificity of the enzyme, there is no risk of overdigestion if the incubation time is prolonged. The enzyme is active at pH 5.5 to 9.0 and does not require reducing conditions or co-factors for activity. The optimal activity is at 37°C, but the digestion can also be performed at room temperature using an increased incubation time.

IgMBRAZOR is expressed in E. coli. The enzyme contains a His-tag and has a molecular weight of 38 kDa.

Unit Definition

One unit IgMBRAZOR digests ≥90% of 1 µg human myeloma IgM when incubated in PBS (10 mM sodium phosphate, 140 mM NaCl, 2.7 mM KCl), pH 7.4 at room temperature for 20 minutes.

Content and Storage

IgMBRAZOR Lyophilized is reconstituted by the addition of water. No preservatives added. After reconstitution, the enzyme is stable for 1 month at +4-8°C. The product is shipped at ambient temperature, and should be stored at -20°C upon arrival.

Yes, IgMBRAZOR and PNGase F can be used in combination if the reaction is performed under non-denaturing conditions. However, for complete deglycosylation of IgM using PNGase F denaturing conditions may be required and it is then recommended to perform the IgMBRAZOR digestion prior the PNGase F deglycosylation.

It is possible to use IgMBRAZOR with low concentrations of IgM. However, this will lead to a decreased reaction rate, so for IgM concentrations below 0.5 mg/mL it is recommended to increase the amount of added enzyme and/or increase the incubation time. There is no risk of overdigestion and the incubation time can be prolonged to overnight if necessary.

Yes, IgMBRAZOR and FabRICATOR can be used in combination.

The enzyme is active at pH 5.5 to 9.0, tolerates up to 300 mM NaCl, and does not require reducing conditions or co-factors for activity. PBS and TBS buffers are both compatible. Optimization may be required if buffers other than the recommended are used.

IgM from some species of monkey may also be digested by IgMBRAZOR.
