FabULOUS Fab kit
Use FabULOUS Fab kit to generate purified and homogenous Fab fragments. The kit involves digestion of mouse IgG with FabULOUS and purification of the Fab fragments on LC-Kappa (murine) specific affinity spin columns.
FabULOUS Fab kit contains one vial of FabULOUS enzyme with 2000 units for digestion of up to 2 mg mouse IgG and 4 LC-Kappa (mur) micro spin columns where each column includes sufficient material to purify up to 0.5 mg IgG.

FabULOUS Fab kit is for rapid and easy preparation and purification of Fab fragments from mouse antibodies. The kit contains FabULOUS enzyme for digestion of mouse IgG and LC-Kappa specific affinity spin columns for purification of the Fab fragments.
FabULOUS enzyme is a unique recombinantly produced cysteine protease that digests antibodies from many species and subclasses in the hinge region producing a homogenous pool of Fab fragments. The enzyme requires reduced condition for activity and reaction is performed in the presence of 50mM Cysteine at neutral pH.
The CaptureSelect LC-Kappa (mur)* affinity matrix recognizes the LC-Kappa domain of murine IgG antibodies, which enables purification of Fab fragments.
FabULOUS Fab kit is shipped on ice.
FabULOUS 2000u should be stored at -20°C upon arrival.
CaptureSelect LC-Kappa (mur) resin* in MicroSpin columns should be stored at +4-8°C.
After reconstitution of FabULOUS enzyme, it is stable for 1 months at +4-8°C.
One unit digests ≥ 95% of 1µg human or mouse IgG when incubated in 10 mM Tris, 150 mM NaCl pH 7.4 at 37°C for 1 hour.
Purified and homogenous Fab fragments can be used in many applications, including:
- Study monovalent binding of Fab fragments to the antigen
- Elimination of Fc-mediated effector functions
- Reduced unspecific binding from Fc interactions
