The Immobilized SialEXO Microspin columns contain SialEXO covalently coupled to agarose beads, for desialylation of up to 0.5 mg native glycoproteins in 30 minutes without any residual enzyme in the final preparation.

Hydrolyzes sialic acids (α2-3, α2-6 and α2-8) on native glycoproteins or released glycans.

Detailed Information

Immobilized SialEXO is a resin with a mixture of two sialidases covalently coupled to agarose beads for complete removal of sialic acids (α2-3, α2-6 & α2-8, linked) on O- and N-glycosylated proteins. The enzymes in SialEXO are derived from Akkermansia muciniphila and expressed in E. coli.

Desialylated proteins are generated without any residual enzyme in the final preparation. The glycoprotein sample is incubated with the resin for 30 minutes at room temperature and the desialylated glycoproteins are then easily collected by a centrifugation step. The recommended buffer is 20 mM Tris pH 6.8. The protocol may need optimization regarding buffer compatibility and incubation time for individual glycoproteins.

Storage and Stability

Supplied in 20% ethanol, shipped cold, and should be stored at +4-8°C upon arrival. Do not freeze the product!

0.1 ml of settled Immobilized SialEXO agarose resin hydrolyzes sialic acids from > 90% of 0.5 mg C1 inhibitor in 30 min at room temperature in PBS pH 7.4 as monitored by ion exchange chromatography.
