FucosEXO Enzyme
FucosEXO is a mix of α-fucosidases for efficient hydrolysis of α1-2, α1-3 and α1-4-linked fucose on N- and O-glycosylated proteins or free oligosaccharides. The enzymes in FucosEXO are expressed in E. coli with a His-tag, and have molecular weights of 87 kDa and 64 kDa. FucosEXO hydrolyzes fucose on glycoproteins under native conditions and displays high activity in a pH range from 6 to 8, without the need for co-factors or additives. The enzyme is provided lyophilized in 2000 unit vials or immobilized on agarose beads in a micro spin column format for processing of up to 0.5 mg of glycoprotein without enzyme in the final preparation.
Unit Definition
One unit of FucosEXO hydrolyzes fucoses from ≥90% of 11 nmol 2'-fucosyllactose and 4.5 nmol 3-fucosyllactose when incubated in 20 mM Tris pH 6.8 at 37°C for 30 min.
Content and Storage
FucosEXO is supplied lyophilized in TBS pH 7.6, with no preservatives added. FucosEXO is shipped cold and should be stored at -20 °C upon arrival. After reconstitution FucosEXO is stable for at least 1 month at +4-8 °C.
Immobilized FucosEXO Microspin columns contain sufficient material each to remove fucoses from 0.5 mg glycoprotein. The resin is supplied in 20% EtOH with no preservatives added. Immobilized FucosEXO is shipped cold and should be stored at +4-8°C upon arrival. Do not freeze the product!
