Multi-Attribute mAb Variant Characterization with CE-MS and FabRICATOR® Digestion

Scientists at Aalen University and their collaborator present a detailed methodology for the rapid and thorough characterization of monoclonal antibody (mAb) variants using capillary electrophoresis-mass spectrometry (CE-MS) at the subunit level. mAbs are an important class of therapeutic molecules for treating various diseases, including cancer and autoimmune disorders. mAbs are large, complex molecules and can undergo a range of post-translational modifications (PTMs), which may affect their biological activity, stability, and therapeutic efficacy. Comprehensive characterization of these modifications is crucial to ensuring the safety and quality of therapeutic mAbs.

Unravelling Structural Conformers within a Therapeutic Multispecific mAb using FabALACTICA


Scientists at the University of Strasbourg and Sanofi describe the global structural characterisation of a trispecific antibody which exhibits two distinct structural isoforms when analyzed by size-exclusion chromatography (SEC). FabALACTICA proved to be a valuable tool in this study as above hinge digestion of this trispecific antibody allowed for middle-level characterisation of the isomeric Fab regions and facilitated cyclic-ion mobility (cIM) differentiation of the isomers, which was not possible in the intact molecule without prior SEC separation.

New reference of FabRICATOR in antibody based PET imaging

To develop an antibody based PET imaging for detection of amyloid beta in mouse models, Sehlin and colleagues  at the Rudbeck Laboratory in Uppsala, Sweden, used Fabricator to generate F(ab’)2 fragments. In the study, published in Nature Communications, Sehlin et al. generated F(ab’)2 fragments using FabRICATOR of an amyloid beta specific antibody and conjugated this to a transferrin receptor antibody for delivery across the blood brain barrier.
