At the 59th ASMS conference last week Ellis and co-workers at ImmunoGen compared several deglycosylation enzymes on different species of antibodies and antibody drug conjugates (ADC). The poster presented compared activity and time for complete N-linked deglycosylation of antibodies. The enzymes used in the study were PNGase F, Endo F3 and IgGZERO. On all antibodies IgGZERO showed highest activity requiring shortest incubation time 30 minutes – 4 hours compared to 24 hour for other enzymes. Also, IgGZERO was the only enzyme that completely deglycosylated all the antibodies tested.
