FabRICATOR (IdeS)

The extraordinary specificity of the FabRICATOR enzyme allows for precise digestion of IgG at one single site in lower hinge of all human IgG sub classes. IgGs are the only known substrate for this enzyme. Reaction conditions using the FabRICATOR enzyme does not require additives, denaturing agents or extreme pH. Additionally, reaction is completed within 30-60 minutes at neutral pH minutes thereby eliminating artifacts induced by sample preparation.

FabRICATOR has been cited in multiple scientific papers during the last 3 years for it's ability to simplify antibody analytics using many different technologies including HPLC, LC/MS, cIEF. GlycINATOR (Endo S2)

GlycINATOR removes N-Glycans from the Fc domain with high specificity for IgGs. The Fc-glycan is digested just after the first GlcNAC leaving one GlcNAC and the fucose attached to the amino acid. GlycINATOR removes common structures (G0, G0F, G1...) including high mannose and hybrid type glycans. Reaction is performed in common buffers without additives or detergents at neutral pH. Reaction is performed in 30 minutes without extensive method development or optimization needed. Learn more about GlycINATOR.

IgGZERO removes N-Glycans from the Fc domain with extremely high specificity for IgGs. Actually, the Fc domain of IgG is the only known substrate for the enzyme and it shows the same high specificity for Fc-fusions. The Fc-glycan is digested just after the first GlcNAC leaving one GlcNAC and the fucose attached to the amino acid. IgGZERO removes common structures (G0, G0F, G1...) but not high mannose and it has very limited activity towards hybrid type glycans. Reaction is performed in common buffers without additives or detergents at neutral pH. FabULOUS digest many different species of IgG in the upper hinge region of IgG. Digestion is performed at neutral pH under reducing conditions. Human IgG1 is primarily digested as the position depicted above. FabULOUS digests most IgGs using a rapid 60 minute digestion protocol. Learn more about FabULOUS
