NIMT®FeOfection|PURPLE is a new transfection agent for siRNA delivery developed by Genovis, featuring consistent high transfection efficiency and very low cell cytotoxicity. It is based on lipid coated iron oxide nanoparticles. The particles consists of solid iron oxide cores that are coated with a specific cationic lipid formulation to facilitate siRNA binding, and cellular uptake. With NIMT®FeOfection transfection reagents effective gene silencing is reached in a wide range of cell lines including suspension cells, mesenchymal stem cells as well as adherent standard cell lines.

Gene Silencing Using NIMT®FeOfection|PURPLE for siRNA Delivery

NIMT®FeOfection|PURPLE is easy and fast to use. Simply mix particles and siRNA, incubate 15-20 min in room temperature and then add to cells. Incubate the cells 24-48 hrs and then read knockdown. NIMT®FeOfection|PURPLE can be used with both forward and reverse transfection.

HEK293 cells are efficiently silenced when using NIMT®FeOfection|PURPLE for delivery of siRNA. Reducing the levels of KIF11 (Eg5) in cells results in mitotic arrest. The cells appear rounded rather than flat and eventually they die. In figure 1a knockdown of Eg5 in HEK293 cells is clearly visualized. In figure 1b cells treated with negative control siRNA show normal phenotype.

Transfect with Low siRNA Concentration – Minimize Off-Target Effects

Lowering the siRNA concentration reduces the risk of unwanted off-target effects in the cell. With NIMT®FeOfection transfection reagent a high gene silencing effect is maintained when transfecting with a siRNA concentration of 5 nM.



 



Gene Silencing in Suspension Cells

Good gene silencing is achieved in suspension cell lines K562 and Jurkat when using NIMT®FeOfection|PURPLE as transfection agent. Extending the transfection time to 48h increases the level of gene silencing.
  



Gene Silencing in hMSC

Excellent knockdown is reached in Mesenchymal Stem cells at both the mRNA (>95% knockdown) and protein (70% knockdown) level when using either NIMT®FeOfection|PURPLE or NIMT®FeOfection|PINK for delivery of the siRNA. 
  



Gene Silencing in Standard Adherent Cell Line

Close to 100% knockdown with a low effect on cell viability is attained in 786-O cells transfected with siRNA targeting gapdh.
