FabRICATOR^®Kit

FabRICATOR^® utilizes the unique specificity of a novel proteolytic enzyme that cleaves IgG at the same place in the hinge region creating pure F(ab')2 fragments without any further degradation. The FabRICATOR^®Kit provides not only precise antibody fragments, but also simple and rapid purification of F(ab')2 fragments in a single step utilizing magnetic particles. With the FabRICATOR^®Kit IgG is cleaved and purified F(ab’)2 fragments are available in less than 1h. 

Production of F(ab')2 Using FabRICATOR^®Kit

The FabRICATOR^®Kit contains:

• One vial of Reagent 1: FabRICATOR^®
• One vial of Reagent 2: Magnetic particles carrying protein A
• One test tube holder with a magnet for magnetic separation of Fc fragments

Procedure: 

1. Mix IgG with Reagent 1 in a test tube and incubate 30 min at 37°C. 
2. Add Reagent 2 and incubate 15-30 min at room temperature.
3. Put the sample on the magnet, let the particles separate, and transfer the supernatant into a clean reaction vessel. The supernatant contains only F(ab’)2 fragments.

What FabRICATOR^® Kit to Buy

• FabRICATOR^® Test Kit contains enough Reagent 1 and 2 to cleave and purify 50 µg of IgG and a test tube holder with a magnet.
• FabRICATOR^® Kit 2000 u contains enough Reagent 1 and 2 to cleave and purify 2 mg of IgG and a test tube holder with a magnet.
• FabRICATOR^® Kit 5000 u contains enough Reagent 1 and 2 to cleave and purify 5 mg of IgG and a test tube holder with a magnet.
• FabRICATOR^® Refill Kit 2000 u and 5000 u contains enough Reagent 1 and 2 to cleave and purify 2 mg and 5 mg of IgG, respectively.

NOTE! The refill kits do not include the test tube holder with a magnet that is inluded in the FabRICATOR^® Test Kit, FabRICATOR^® Kit 2000 u and FabRICATOR^® Kit 5000 u. The test tube holder with a magnet is necessary for the kit to work. If you already have purchased one of the FabRICATOR^® Kits and you have a holder, you can purchase one of the Refill kits.
